Recombinant monoclonal antibody to Integrin Integrin alpha 5 beta 1. Volociximab (also known as M200) is a chimeric monoclonal antibody jointly for treatment of a variety of advanced solid tumors. It binds to and inhibits the functional activity of α5β1 integrin.
It is thought to reduce metastases. Early results show potential in renal cell cancers.
Figure 1 Testing antibody cross-reactivity to human α5β1.
IIA1, the mouse parent antibody of volociximab, which recognizes only human integrin, anti-mouse CD31, which stains mouse vessels, and pooled rat IgG are shown as controls.

Figure 2 Representative antibodies were compared by ELISA for binding to human and murine α5β1-Fc or irrelevant Fc fusion proteins.
A subset of competitive antibodies cross-reacted with human integrin.

Figure 3 Binding of volociximab to integrin α5β1 and inhibition of integrin binding to fibronectin.
A dilution series of volociximab was incubated with immobilized human, mouse or rabbit integrin α5β1- Fc fusion proteins and bound volociximab was detected using an anti-human κ-chain specific antibody.

Figure 4 VX2 prevention study. Thirty rabbits were inoculated with subcutaneous tumors on the left hind limb.
Volociximab-treated rabbits generally grew fewer tumor-associated vessels than vehicle-treated rabbits

DrugMonitor™ Anti-Volociximab Antibody (VS-1224-YC1312)Volociximab is a chimeric monoclonal antibody developed for treating various advanced solid tumors. The DrugMonitor™ Anti-Volociximab Antibody (VS-1224-YC1312) is an anti-drug antibody (ADA) against Volociximab. This drug-based antibody is raised in mice immunized with the Volociximab. The anti-Volociximab antibody may be used in ELISA, pharmacokinetics (PK), and pharmacodynamics (PD) analyses, or serves as a reference standard in ADA assays. It also is an excellent tool for therapeutic drug monitoring, allowing to evaluate the drug efficacy and determine the drug concentration of the Volociximab in samples.
